Study of the bone dynamics of an implant surface coated with odanacatib through wettability, topographical, biomechanical, computed microtomography, histomorphometric, biocompatibility, culture of cells and immunohistochemistry

Abstract

Currently, studies found in the literature promote the success of osseointegration related to surface treatment of titanium implants. Addressing the challenges of finding chemical components that promote changes in favor of the bone neoformation process. However, there is still a need to improve the technology of these materials for early bone regeneration. Thus, the objective of this work is to evaluate the dynamics of the bone response of a surface modified by acid etching, modified by acid plus Body Fluid Solution (SBF), and modified by the active ingredient Odanacatib, at a concentration of 0.86mg / L, by the biomimetic method in a standardized rat tibia model. For this, 144 male rats (Rattus Copernicus Albinus Wistar) and 288 commercially pure titanium implants of 1.6 in diameter and 3.0 mm in height will be available, 96 with double acid etch surface (GC), 96 with surface modified by double acid etching, and SBF (GX) 96 with modified surface based on Odanacatib (GO) by the biomimetic method of surface modification that will be installed on the tibiae of rats. The periods of euthanasia for the collection of tibias will be 15 and 40 days. For an analysis of cell culture used, 18 male Wistar rats (Rattus Copernicus Albinus Wistar) and 108 titanium disks of 0.5 cm in diameter and 0.3 cm in height will be used for 24 wells, 36 with double surface acid etching (GCC), 36 with surface modified by SBF (GXC) and 36 with surface modified based on Odanacatib (GOC) by the biomimetic method of surface modification. For a subcutaneous analysis, 48 rats (Rattus Copernicus Albinus Wistar) will be exposed and 96 titanium discs of 0.5 cm in diameter and 0.3 cm in height will be used, 32 with double acid etch surface (GCS), 32 with surface modified by SBF (GXS) and 32 with surface modified based on Odanacatib (GOS) by the biomimetic method of surface modification that will be installed 2 discs per animal in the subcutaneous tissue in the extreme regions of the animal's back, in which tissue collection subcutaneous will be 3 and 7 days. The specimens will be decomposed to the analysis of wettability, topography, cell culture, biomechanics, computerized microtomography, histology, histometry, and immunohistochemistry. Finally, the quantitative data of the studied groups will be collected through a statistical analysis, adopting the significance level p <0.05. (AU)
