Abstract

The aim of this study will be to evaluate the biological effect of bioactive cements on stem cells from human exfoliated deciduous teeth (SHED). The samples will be obtained by means of primary culture of SHED. The pulp tissue will be removed and stored in DMEM supplemented with 20% Bovine Fetal Serum. The cements will be prepared at a concentration of 1mg / mL MEMa with 10% fetal bovine serum according to the experimental groups: Group 1: BBio, Group 2: Bio-C Repair, Group 3: MTA HP Repair. Cell viability experiments will be carried out using the MTT method and cell migration. The alkaline phosphatase activity test will be performed, and the alizarin red will be used to assess the formation of the mineral nodule. The gene expression of odontogenic markers will be evaluated by the RT-PCR method, and the production of DMP-1 and DSPP will be quantified. In the statistical analysis, the Shapiro-Wilk normality test will be applied. For data showing normal distribution will be performed, the analysis of variance of repeated measures test, followed by the Tukey test. For data that do not have a normal distribution will be performed the Friedman test and the Kruskal-Wallis test. For all tests, the level of significance adopted will be 5%. (AU)
