Abstract

Mice models for polycystic ovary syndrome and controls will be subjected to high calorie diet and supplementation or not with sodium butyrate for 40 days. The animals will be evaluated for their total weight, insulin resistance and resting energy expenditure at the beginning and at the end of supplementation. After euthanasia, blood, liver, pelvic, femur, tibia and visceral, subcutaneous and interscapular adipose tissues will be harvested. The serum level of leptin and adiponectin will be assessed. The adipose tissues will be weighed and processed for histological evaluation and expression of leptin, ghrelin, adiponectin and uncoupling proteins 1 (UCP1) and PRDM 16. The femurs bone marrow will be collected for expression of adiponectin and UCP1. The tibiae will be stained with osmium tetroxide to assess bone marrow adipose tissue by microtomography. Stool samples will be collected at the beginning and at the end of the study to assess the diversity of the intestinal microbiota. (AU)
