Nanoparticles coated with glutathione and albumin for applications in nanomedicine

Abstract

Nanomaterials can be designed with defined sizes, shapes and compositions and utilized in a wide range of biomedical applications. Advances in synthesis and nanoengineering allow the preparation of increasingly sophisticated nanomaterials that can perform increasingly complex biological functions. However, among those nanomaterials with prominent in vivo performance and approved for use in humans, most exhibit a simpler architecture. It is widely accepted that fundamental studies on nanomaterials and their interactions with biological systems are urgently necessary so that we can understand and eventually design nanomaterials with controlled biological behavior. Herein, we propose to study three models of nanoparticles (NPs) of extremely simple architecture and the interactions of these particles with selected biological systems (fetal bovine serum, human plasma and cells in culture). 50 nm silica NPs, 1.2 nm gold nanoclusters, and 5 and 30 nm colloidal gold NPs will be coated with albumin and glutathione (GSH) in order to obtain inert, biocompatible surfaces that will be resistant to biomolecular adsorption. The silica NPs will be coated with native and cationized albumin. The nanoclusters will be synthesized from GSH and two derivatives, GSH monoethyl ester and GSH diethyl ester. The colloidal gold NPs will be coated with GSH monoethyl ester. The NPs will be evaluated for their colloidal stability, ability to resist protein adsorption, blood biocompatibility (focus on coagulation), and cellular internalization in vitro. The NPs will be also functionalized in order to demonstrate their ability to recognize specific cell receptors in biological media. These fundamental studies will provide a greater understanding of the behavior of NPs coated with albumin and glutathione in biological systems. Our results will be important for future applications of these NP models with recognized clinical potential. (AU)
