Global Risk Assessment of Malignant Transformation in Oral Potentially Malignant Disorders (OPMDs): Technical-Scientific Integration and Sustainability

Abstract

This project aims to develop and validate a predictive model to assess the risk of malignant transformation in oral potentially malignant disorders (OPMD), based on the integration of clinical, histopathological, and cytometric data. By employing machine learning and flow cytometry, the proposal seeks to overcome the limitations of isolated biomarkers, offering a robust, scalable, and clinically applicable tool. Data collection will be prospective and longitudinal, and model performance will be evaluated through cross-validation and comparison with the Newcastle Nomogram. The project proposes the development of a structured digital platform to consolidate these data, with a focus on clinical applicability, data security, and analytical accuracy. Beyond the technical-scientific dimension, the implementation process will be guided by Implementation Science, using the CFIR, EPIS, and RE-AIM frameworks to ensure institutional adoption, long-term sustainability, and real-world impact. This proposal is aligned with the Brazilian National Agenda for Health Research Priorities (ANPPS) and holds strong potential for adoption in both public and private healthcare systems. (AU)
