Abstract

Leptin is an adipocyte-derived hormone that acts to reduce food intake and increase energy expenditure by binding to specific receptors in the hypothalamus. Variants of leptin (LEP) and leptin receptor (LEPR) genes have been associated with increase of plasma leptin and obesity-related phenotypes. The aim of this study is evaluate the influence of LEP and LEPR variants on body composition and plasma markers for obesity in the Brazilian population. Anthropometrics parameters, body fat, and biochemical parameters will be evaluated in 300 non-related individuals. Peripheral blood samples will be used for DNA extraction and leptin, soluble leptin receptor and lipid profile analyses. The regions containing the variants LEP (G-2548A) e LEPR (Lys109Arg, Gln223Arg e Lys656Asn) will be amplified by polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP). We expect that the results driven from the analyses may contribute to the knowledge of the role of LEP and LEPR variants on the development of the obesity. (AU)
