DNA damage induced by metal ions from orthodontic therapy: in vivo and in vitro analyses

Abstract

Nowadays, orthodontic therapy plays an important role with respect to restablish stomatognathic system as fas as contributing for esthetic harmonization to patients. Nevertheless, there are few studies that clarify if corrosion products from orthodontic therapy are able to induce genetic damage so far. Herein, the aim of this study will be to evaluate the putatitve genetic damage induced by metal ions discharge from orthodontic therapy by means of single cell gel (comet) and micronucleus assays in vivo and in vitro. For this, the following parameters will be evaluated: 1) DNA damage in buccal cells by the micronucleus assay following nichel-titanium arches and inox steel appliances in vivo; 2) genotoxicity in ordinary human lymphocytes exposed to corrosion eluates from orthodontic appliances by single cell gel (comet) assay in vitro; 3) mutagenicity in human ordinary lymphocytes exposed to corrosion eluates from orthodontic appliances by the micronucleus assay in vitro.
