Tripanocidal activity evaluation of crude extract and fractions of Piperaceae plants

Abstract

Trypanosoma cruzi is the ethiological agent of Chagas disease and transmitted to human by triatomine bugs or through blood transfusion, which affects 16 to 18 million people in Latin America. The therapy continues partially inefficacious despite the efforts of the researchers. In the current literature, there are many investigations on the tripanocida activity for a great variety of natural extracts, especially those from vegetables, or isolated compounds and half-synthetic ones. These analyses can be made through a process of counting of parasites on the microscope, which demanding great time, intensive work in the laboratory and the presence of an unique observer; or using colorimetric methods, as MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] reduction in formazan that is easier and faster application. This colorimetric assay has successfully been used with mammalian cells and also with some protozoa. So, this project focus to evaluate the tripanocida activity of extracts and soluble fractions of plants of the Piperaceae family in epimastigotes forms of Trypanosoma cruzi by using the colorimetric method as an efficient faster test. The samples, which present significant activity, will be also analyzed in phase contrast microscopy for evaluation about the morphologic variations of the parasites. (AU)
