Evaluation of serological and contamination profile of Pasteurella multocida in swine herds

Abstract

Pasteurella multocida is an important pathogen that infects swine, causing progressive atrophic rhinitis, pneumonia, pleuritis and septicemia. The implantation of control and prevention strategies against this infection become necessary the knowledge about the agent spreading in natural conditions and in swine herds with different management systems. The present study proposes the determination of immune status of animals against the agent in different ages through ELISA in nine herds where the P. multocida is present and different control and prevention programs are used. The serological profile will be accompanied by elimination profiles obtained through PCR. It will be used immunoenzymatic tests to detect antibodies against P. multocida and dermonecrotic toxin, related to progressive atrophic rhinitis. The PCR will carry at the same way to search the DNA of agent and the gene codifying the dermonecrotic toxin. The swine herds will be selected at first, by identification of agent and lesions at slaughterhouses. Among the positive herds selected will be chose three herds without vaccination program and low use of antimicrobials, three herds using commercial vaccine at sows and three herds using commercial vaccines in sows and piglets. Blood samples and tonsils swabs of 14 animals from the following ages will be collected: 20 days, 40 days, 60 days, 90 days, 110 days and 140 days. Blood samples, tonsil swabs and colostrum samples from four sows from each herd will be collected also. A total of ninety-two samples from each herd will be examined through ELISA and PCR. From obtained results will be possible to get information about the spreading pattern of agent in swine herds and the best option of prevention and control in rhinitis and pneumonia outbreaks. (AU)
