The effect of b-glucan and vitamin C on the specific and non specific immunological response in pacu (Piaractus mesopotamicus)

Abstract

The aim of the present work will be to evaluate the effect of glucan and high level vitamin C association (ascorbil poliphosphato) in diet, on non specific and specific response in pacu (Piaractus mesopotamicus) submitted to a Aeromonas hydrophila infection. 192 juvenile fish, with 200 to 300g weight, will be distributed in experimental box with 300 L (4 treatments, 6 repetition, 8 fish/box) and feed for four weeks with the experimental diets (0,0% de 1,3-glucan/kg ration and 200mg/kg ration of vitamin C); treatment 1 (0,5% de 1,3-glucan/kg ration and 200mg/kg vitamin C), treatment 2 (0,5% 1,3-glucano/kg ration and 1000mg/kg vitamin C) and treatment 3 (0,5% 1,3-glucano/kg ration and 2000mg/kg vitamin C). After this period, the fish will be to sample (6 weeks) and submitted to Aeromonas hydrophila challenge in the intraperitoneal cavity. The fish continue receiving the control diet for two weeks and blood sample again (after challenge). Each time, blood and renal tissue will be sample to number of erythrocytes determination, hematocrit, hemoglobin concentration, mean corpuscular volume (MCV) and white blood cells), total protein and non specific response (lysozyme concentration and activity, complement activity and burst) and specific response (antibodies assay). The experimental design will randomized entirely by using factorial scheme 4 x 3 (treatments x sample time). The results will performe ANOVA. When the ANOVA identified significant difference among the groups comparison among means were performed with Duncan´s multiple range test. Significant differences between groups and the treatments were accepted at P<0.05. (AU)
