Abstract

Choriocarcinoma is a rapidly invasive epithelial neoplasia that is frequently associated with metastasis. Although its parental origin has already been established in a few molecular reports, there is no large-scale study on prognostic molecular markers. Objectives: The purpose of this work is to: 1) assess genetic molecular features in a series of choriocarcinomas; 2) determine the parental tumor origin (gestational or non-gestational); 3) identify genomic imbalances by comparative genomic hybridization based on arrays (aCGH) in the primary tumor and its metastasis; 4) evaluate the correlations between genomic alterations and clinicopathological outcomes. Methods: This study will include 30 patients under treatment at the Center of Trophoblastic Diseases, Botucatu Medical School Hospital-São Paulo State University-UNESP between 1995 and 2010. Choriocarcinoma samples matched with peripheral blood samples (5mL) from the patient and her partner will be used for genetic parental analysis using polymorphic microsatellite markers. Choriocarcinoma clinical development will be evaluated based on hCG measurements obtained during at least one year of follow up. Genomic imbalances will be evaluated in choriocarcinomas and their metastases (when available) by aCGH. Fluorescence in sity hybridization (FISH) and/or immunohistochemistry will be performed in a large series of choriocarcinomas and their metastases in a tissue microarray in order to confirm the relevance of the putative molecular markers.
