Effects of equine inflammation on hepcidin expression and plasma iron concentration

Abstract

Equine serum or plasma iron concentration drops quickly during inflammation, and its quantification is a reliable procedure to diagnose systemic inflammatory processes. Accumulation of iron inside macrophages and reduction of the intestinal absorption of this element causes hypoferremia during systemic inflammatory process. These mechanisms are mediated by the hepcidin, a 25 amino acid peptide synthesized mainly by the hepatocytes in response to iron stores and inflammation. The higher hepcidin expression during inflammation restricts the iron availability, necessary for pathogenic microorganism growth. This increase in hepcidin expression can be particularly useful during initial periods of inflammation, before adaptive immunity occurs. The hepcidin sequencing has already been performed in human, rat, mouse, swine, fish and dog; however hepcidin in horses has not been sequenced yet. Here we propose to sequence the responsible gene for hepcidin codification in equine, to verify its expression in different tissues and to evaluate if inflammatory response induction decreases plasmatic iron concentration and check if this was preceded by an increase in hepcidin expression.
