Abstract

Indoleamine 2,3-dioxygenase (IDO) is an enzyme involved in the cataboiism of tryptophan and by limiting the offer of this essential amino acid, impairs proliferation of alloreactive T cells, thus helping preventing rejection of the conceptus during pregnancy. Several studies have been conducted on the role of IDO on maternal-fetal tolerance in human and murine placental tissue. Changes in IDO expression and activity are related to fertility problems in humans and lower IDO activity was found in women who had recurrent abortions. Reproductive hormones also play a part on the fetus survival process, like progesterone that blocks mitogenic stimuli for T cells proliferation, modulates antibody production, favors IL-10 production, etc, and estrogen that may, depending on its concentration, change Th1 or Th2 response in gestation. Aiming understanding the possible relationship between these hormones and indoleamine, IDO expression behavior and cell cycle of cultured placenta and fetuses cells from Balb-C mice and Wistar rats will be evaluated by flow cytometry after the addition of progesterone, estrogen, γ interferon, tryptophan and 1 -methyl-tryptophan to the culture.
