Effect of season on oocyte quality and in vitro embryo production in Holstein cows (Bos taurus) of different categories

Abstract

The use of in vitro embryo production (IVP) technique to improve the reproductive capacity of the domestic animals has been substantially important for animal production. However, the IVP is still very inefficient for high producing Holstein cows and influenced by many factors. The environmental conditions to which the animals are submitted to is one important factor to be considered. Moreover, the category of the animal used as oocyte donor can also be decisive for IVP success. Thus, in the present proposal, the effect of the season of the year (warm and humid x fresh) will be studied in different animal categories (high-producing lactating cows, repeat breeder lactating cows and heifers) in a commercial Holstein (Bos taurus)herd. The main objective of the present experiment is to determine the best season and categories of Holstein females to be submitted to follicular aspiration for in vitro embryo production, based on physiological and molecular analysis. First, the physiological parameters, the recovery rate and the rate of viable oocytes recovered by in vivo ovum pick-up (OPU) technique will be analyzed. After, the quality of these recovered oocytes will be evaluated by the expression of genes related to heat stress (HSP70 e HSP90) and apoptosis (Bax, Bcl2 e ITM2B) and by quantitative mitochondrial DNA (mtDNA). Additionally, the rates related to in vitro embryo development after in vitro fertilization and the blastocysts quality ( total number of cells and frequency of apoptotic nuclei) will be evaluated.
