In vitro embryo production in Bos indicus and Bos taurus heifers supplemented with diet different levels of energy

Abstract

The objective of this study will be to evaluate the effect of the diet with different levels of energy in oocytes quality the in vitro embryos yield Bos indicus and Bos taurus heifers and to correlate it with the metabolic and hormonal profile. In this study, 30 heifers [15 Bos indicus (Gir) and 15 Bos taurus (Holstein)] will be distributed randomly in 3x2 factorial design: 1) Low energy indicus - the Gir heifers (Bos indicus) received 90% maintenance diet, 2) Low energy taurus - the Holstein heifers (Bos taurus) received 90% maintenance diet, 3) Moderate energy indicus - the Gir heifers (Bos indicus) received 150% maintenance diet, 4) Moderate energy taurus - the Holstein heifers (Bos taurus) received 150% maintenance diet. 5) High energy indicus - the Gir heifers (Bos indicus) received 200% maintenance diet, 6) High energy taurus - the Holstein heifers (Bos taurus) received 200% maintenance diet. The animals will be submitted the twelve (12) ovum pick-up (OPU) with intervals 14 days. The complex cumulus-oocytes quality and the number of follicles in ovary [small (<6mm), medium (6-9mm) and big (>9mm)], the diameter of the two biggest follicles will be analyzed. In vitro embryos yield will be evaluated the complex cumulus-oocytes quality, recovery rate, cleavage rate and in vitro production of embryos. In the metabolic evaluation, non-esterified fatty acids (NEFA), urea, ²- hydroxybutyrate, cholesterol and glucose will be analyzed. In the hormonal evaluation, the insulin, IGF-1 and leptin concentrations will be analyzed. The dependent variables of normal distribution (parametric) will be expressed in mean and standard error (mean+-SEM) and analyzed repeat measures in time using the proc MIXED of SAS and the one of abnormal distribution (nonparametric) by NPAR1WAY WILCOXON procedure of SAS. The binomial and discrete variables will be analyzed by procedure GLIMMIX of SAS. (AU)
