Abstract

Vitamin D receptor is expressed in several tissues, and when activated modulates gene expression in target cells in the kidney, pancreas, skeleton, adipocytes and vascular smooth muscle cells. Recent evidences linked vitamin D nutritional status with the components of metabolic syndrome. Aim- To investigate the relationship between vitamin D nutritional status and vitamin D receptor (VDR) polymorphisms with the components of metabolic syndrome in adults and elderly. To establish a cut off for vitamin D serum concentration for extra bone outcomes. Methods - Cross sectional study, including 500 free living individuals older than 18 years. The present study is a subset of the "Vitamin D nutritional status and its relationship with the components of metabolic syndrome in adults and elderly of São Paulo" FAPESP no 07/52420-2). In summary, nutrient intakes was evaluated by a 24hr food record, body composition by bioelectrical impedance and waist and hip circumference. Vitamin D nutritional status was evaluated by serum 25(OH)D3, intact parathormone, vitamin D intake and sun exposure. The presence of metabolic syndrome was classified by the NCEP-ATP III criteria. Blood pressure, total cholesterol and triglicerides, glucose and insulin was also measured. In the present analysis, VDR polymorphisms will be performed by polymerase chain reaction (PCR) including specific digestion enzymes. For statistical analysis the Hardy-Weinberg equilibrium test, as well ANOVA and/or ANCOVA tests will be performed in order to investigate the differences between genotypes. The ROC curve will be constructed for extra bone serum 25(OH)D concentration adequacy. Expected results - To associate the polymorphisms of VDR gene with the components of metabolic syndrome and to propose an adequate cutoff for extra bone serum 25(OH)D concentration, based in specific criteria for our population.
