Abstract
Human breast and oral cancer constitute important health care problems in Brazil. A complex pattern of genetic and epigenetic alterations are involved in process of tumorigenesis, which often confers to transformed cells a higher proliferative potential, evasion to apoptosis, sustained angiogenesis, and capacity to invade surrounding tissue and metastasize. Extracellular matrix molecules play important role influencing malignant behavior. Our Laboratory has been studying involvement of laminin and its bioactive peptides in salivary gland tumors. We have demonstrated participation of different laminin-derived peptides regulating normal and neoplastic salivary gland cells. This prompted us to study whether these peptides would regulate gene expression in tumor cells. In this project, cells from carcinomas and adenocarcinomas will by treated by laminin-derived peptides C16 and AG73. Cells treated by scrambled peptides will serve as controls. After treatment, cells will be collected for RNA extraction, followed by cDNA synthesis, cRNA synthesis and hybridization with microarray membranes. GEArray Expression Analysis Suite software will be used to normalize and analyze the data. Microarray results will be validated by qPCR, Western Blot and functional assays. (AU)
