Interactomic analysis of hexokinase-VDAC-ANT complex in rat, bovine and chicken neuronal cells

Abstract

The voltage dependent anion channel is the most abundant protein of outer mitochondrial membrane. VDAC controls the metabolite exchange through this membrane and the apoptosis machinery. Interactomics is the study of protein complexes, their interactions and the consequences of these interactions. In our case we will study the interactomics of VDAC and other proteins inside the neuron. Other purpose is to understand the stoichiometry of the complex hexokinase-VDAC-ANT, in chicken, rat and bovine brain. Previous studies from our laboratory have shown a differential expression of VDAC 1 and VDAC2 between these brains. To address this problem, in the present study, we wish to see if the there are differences in the interactions of these proteins in the different brains. Chicken brain mitochondria show the highest VDCA1 expression and the lowest of VDAC2. Other purpose is to see if there is a VDAC1 and VDAC2 differential expression in avian neuronal cells submitted to apoptosis by staurosporin. (AU)
