In vitro evaluation of citotoxicity and genotoxicity of different root canal sealers in cell cultures

Abstract

The successful of endodontic treatment is directly related to an accurate diagnosis, establishing a correct prognosis and treatment plan, as well the use of filling materials that promote good root canal sealing, showing good biological performance, since they are in permanent contact with the periapical tissues and may release toxic components perpetuating chronic inflammation. In this study, will be evaluated the cytotoxicity and the genotoxicity of four root canal sealers avaiable in the market, AH Plus (Dentsply De Trey, Alemanha); Endo Rez (Ultradent, USA); RoekoSeal (Coltene Whaledent, Alemanha); Experimental Root Canal Sealer- based of Copaiba oil-resin (Cop Endo, Brasil). The materials evaluation will be performed in three different times of cure for cytotoxicity: Oh, 12h and 24h. For the genotoxicity test, the cure time of sealers will be only ]2h. Thus, root canal sealers extracts will be prepared. According to the hardness time, the original extracts will be diluted in series and placed in contact with culture of lung fibroblasts of Chinese hamster (V79), for 24h at 37°C temperature. The cell viability will be measured for the MTT test in spectrophotometer and the genotoxicity determined by comet test to evaluate chromosomal damages and through analysis of micronuclei formation. The results of the cell survival rate and the amount of DNA damage will be statistically analyzed for Mann-Whitney-U (p<0,05) test. (AU)
