Analysis of angiogenic factors and COX-2 expression in cartilaginous tumors: clinical-histological correlation

Abstract

Chondrosarcomas are a heterogeneous group of cartilaginous tumors with different clinical and morphological manifestations. The angjogenesis and neoangiogenesis have been related to tumor growth and prognosis and the expression of COX-2 with biological nature. Few studies have evaluated the tumor angiogenesis in cartilage, especially in combination with markers of malignancy. Objectives: immunohistochemical study of cartilaginous tumors regarding: 1) vascular formation and neoformation through CD34 and CD105 markers, 2) the expression of vascular endothelial growth factor (VEGF), 3) the expression of COX-2, and 4) correlate clinical and histological findings of patients. Methods: a retrospective study will be conducted in specimens of cartilaginous neoplasms obtained from the files of the Pathology Department, FCM, UNICAMP. The specimens shall be cut and stained by routine and special staining, for histological examination and graduation, as much as stained by immuno-histochemical method using the following antibodies: anti-CD34, CD105, VEGF and COX-2. The Chalkley method will be used to estimate the mean vascular density. The microvascular density (MVD) of each tumor will also be evaluated, in the so called "hot spot" areas. The relative quantification of the COX-2 immuno-labeled cells will be randomly and blindly assessed on digital images, using the Imagelab program. From the clinical files will be obtained the following information: age, tumor size, radiological aspects, stage presentation, local recurrence / metastasis and time of follow up. Perspectives: This study may contribute to the elucidation of cartilaginous tumors progression and give support to histological diagnosis of low grade malignant tumors. (AU)
