Abstract

Physical exercise evokes an extensive circulatory redistribution that involves changes of tonus in either arteries or veins. Indeed, the exercise possibly modifies the local mechanisms related to nitric oxide (NO), prostanoids and/or endothelin-1 that influence the effects of sympathetic agonists and angiotensin II (Ang II) in veins. This hypothesis was investigated in our preliminary study that aimed to determinate whether exercise modifies the effects of phenylephrine and Ang II in portal veins taken from trained rats. It was verified the exercise enhanced the rat portal vein responses to these agonists. It is also indirectly evidenced that such enhancement of response to phenylephrine and Ang II appears to involve an enhancement of vasocontractile influence of endothelin-1 that supplants the modulation exerted by nitric oxide (NO) and vasodilator prostanoids. Thus, the present project aims to expand this investigation, verifying the exercise effects upon the local expression of endothelin-1and/or receptors ETA/ETB as well as the actions of exogenous endothelin-1 in rat portal vein. Considering that the aforementioned study showed exercise-induced enhancement of phenylephrine and Ang II responses in portal vein but not in vena cava, it will be also investigated the influence of exercise upon other venous beds responses to these agonists. In this manner, such veins will be studied either by organ bath experiments or by quantitative real time RT-PCR. (AU)
