Abstract

The Semliki Forest Virus (SFV) expression system has been utilized for in vitro expression of several molecules of biotechnology interest, and for in vivo expression of medical interest molecules. Known by the ability to infect several cell types, mainly mammalian cells, the SFV has been proposed as a reliable vaccine vector. However, little information is available about its immunologic and adjuvant properties and its utilization is limited for some technologic constraints, as the imprecise viral titration methodology. In this project, infectious but non-replicative recombinant SFV particles will be obtained from plasmids by molecular biology techniques. These particles will bear the genetic information of the rabies virus glycoprotein (RVGP) and delivery it for in vivo synthesis in mice cells. These particles will be able to induce neutralizing antibodies in immunized mice, as already evaluated in preliminary assays. In order to better determination of virus titers, it will be established a new qRT-PCR methodology, based in the quantification of viral RNA in viral lots. The titrated particles will be utilized for mice immunization and the pattern of immune response will be determined, analyzing humoral and cellular responses. Immunized mice will be eventually challenged against rabies virus infection, in order to determine the potency of SFV-RVGP vaccination. (AU)
