The increased demand for recombinat proteins has been addressed the development of new technologies to improve production of safe and efficient biopharmaceuticals from the animal cell cultures. The culture medium used for the maintenance of these systems does not provide the features necessary for cells to grow on plastic culture substrates without the presence of fetal calf serum. However, the use of FCS is not advisable due to the variations found among the lots, the high content of animal protein and the possibility of the presence of infectious agents. For this reason, technological advances led to development of serum free medium, standardize and able to supply the needs of animal cells. Nevertheless, the process of adaptation of cells may be could take considerable time. Thus, this study aims to evaluate the metabolism of cells adapted to serum free medium and gene expression through the use of the Semliki Forest Virus-SFV system (RNA loading). Two genes have been chosen as a target for this study: the rabies virus glycoprotein (RVGP) and nonstructural protein 3 (NS3) of hepatitis C virus (HCV).
