Abstract

Early prediction of Adult T Cell Leukemia (ATL) in HTLV-1 carriers can be difficult before a disease becomes clinically evident. In this study, we aimed to determine the rate of T cell clonality among HTLV-1 asymptomatic carriers with different titers of proviral load (PL) and to establish a criterion for follow up for those who will present higher PL.METHODS:A sensitive SYBR Green real time PCR will be applied on mononuclear cells from a group of 261 HTLV-1 asymptomatic subjects followed in our institute since March 1997. PL will be classified into three categories; low titer (50 copies/1000 PBMCs), intermediate titer (50-100 copies/1000 PBMCs) and high titer (above 100 copies/1000 PBMCs). Approximately 213 samples will be assessed for their T cell receptor rearrangements by etablished TCRg multiplex PCR using primers able to amplify all Vg and Jg regions. The size and profile of PCR products will be determined using GeneMapper v 3.2-computer software. Appropriate statistical methods will be used and data of T cell clonality will be cross-referenced to PL to allow for subsequent association studies. (AU)
