Abstract
The aim of this in vitro study is to obtain standard caries affected-dentine by dentine demineralization process induced by Streptoccocus mutans biofilm (nicrobiological approach). The method validation will be performed by optical coherence tomography, visual inspection, laser fluorescence (DIAGNOdent) and digital intra-oral radiography (DIOR). As all methods do not destruct samples, all tests will be performed in the same specimens. Eighteen human molars will be grounded in the occlusal surface in order to remove enamel and obtain a flat dentin surface. Specimens will be protected with nail varnish in half of the occlusal surface (control), before microbiological challenge (n=6/group) for 3 different period of immersion in the artificial caries solution: 7 (group 1), 14 (group 2) e 21 (group 3) days. At first, artificially induced caries will be visually inspected and photographed, digital radiographs will be taken and diagnodent values will be recorded. Optical coherence tomography images will be obtained and also lesion depth will be recorded. Through this method we will qualitatively analyze by images and a quantitative register of the depth of carious dentin demineralization. The quantitative data obtained will be statistically analyzed and compared, while the qualitative will be described in comparison to the sound dentin (control). (AU)
