S100A4 and osteopontin immunoexpression evaluation in canine spontaneous mammary carcinomas

Abstract

Mammary neoplasms are the most frequent tumors of the bitch; they have their diagnosis taken as a hard task since there are different clinical variants and histotypes, and the neoplasm behavior is unpredictable since there is no standard behavior that can be associated to the diagnosed histotype. Consequently, the search for new cancer biomarkers that allow an early diagnosis and rapid data regarding tumor evolution have been consistently encouraged; this is an emergent field of research that still needs the establishment of new reliable molecular markers. Osteopontin and S100A4 are promising markers for that purpose since these proteins were found to be overexpressend in various primary human carcinomas than in the corresponding nonmalignant tissues. According to that, it is interesting to research osteopotin and S100A4 utility in the monitoring of mammary carcinomas in bitches. In order to evaluate such proteins an immunohistochemical study performed in tissue microarrays will be conducted using 20 normal mammary tissue and 80 mammary carcinoma tissue samples. Primary antibodies will be anti osteopotin, VEGF, CD117, EGFR, cerbB2, p53, p63, CK5, CK8/18, CK14, CK19, vimentin, p-Cadherin, estrogen receptor, progesterone receptor, and Ki67. The results will be compared with clinical and histologic parameters of the cancer patients. The aim of this study is the acquisition of knowledge regarding osteopontin and S 100A4 immunoexpression in the microenvironment and neoplastic cells of canine spontaneous mammary carcinoma; this will help us to actualize ourselves about new potentially useful biologic factors as prognostic biomarkers.
