Effects of bST injection on serum concentrations of glucose, insulin, IGF-1 and progesterone in nonlactating dry cows in different energy balance

Abstract

The objective of this experiment is to evaluate whether energy balance (EB) and/or bST injection (Lactotropin 500mg, Elanco) affect serum concentrations of glucose, insulin, IGF-1 and progesterone (P4) in ovariectomized dry cows with intravaginal P4 releasing device (CIDR® 1,9 g of progesterone, Pfizer Health Animal, Brazil). Fifteen nonlactating, nonpregnat and ovariectomized Gir x Holstein cows (BW = 640 Kg ± 86) will be randomly distributed into two nutritional groups at d-14 of the experiment, one group (n = 5) will receive diet for weight maintenance (WM) and another group (n = 10) will receive diet to weight gain (0,300 kg/day; Positive Energy Balance - PEB). Animals in the WM group more half of PEB group will receive bST (Lactotropin 500mg, Elanco), totaling three experimental groups, (G1) Maintenance with bST, (G2) PEB with bST and (G3) PEB without bST, within 5 cows/group. The animals in PEB will be supplemented individually at 6:00 AM and at 6:00 PM, from d-14 to d-3 with 2 kg of ration per day, and after this period will be provided 4kg of ration of d-2 to d27 in order to gain weight in this group. The animals in WM will be supplemented individually with 1 kg of ration daily at 6:00PM of the d-2 to d27, to minimal loss or weight maintenance. The bST will be applied at d0, d9, d18. Each cow will receive a CIDR device of third use at the adaptation period (d-14 to d-2), and after this period (d-2) each cow will receive a new CIDR that will remain until the end of experimental period (d-2 to d27). Blood samples for glucose, insulin, IGF-1 and P4 will be collect (d0 to d27) at 0h (before supplementation in the morning), 1h and 2h after supplementation, from either the coccygeal vein or artery.
