Abstract

The implantation of bioactive glasses or glass-ceramics represents an important therapeutic strategy aiming to promote bone matrix formation in critical-sized bone defects. When a bioactive glass or glass-ceramic material interacts with biological fluids, changes in surface chemistry and surface topography take place, affecting the interactions of cells with the material surface, resulting in a reduction in cell spreading and altering the labeling pattern for cytoskeletal proteins and integrins. The present study aims to evaluate whether changes in the labeling pattern for actin and tubulin, which take place in osteogenic cells directly interacting with either Bioglass® 45S5 or Biosilicate® glass-ceramic, are due to a reduction in mRNA levels and/or in protein expression. At days 3 and 7 of primary osteogenic cell cultures, actin and tubulin mRNA levels and protein expression will be determined by means of Real-time PCR and enzyme immunoassay (ELISA), respectively.
