Abstract

The diabetes mellitus (DM) represents a group of metabolic disorders has in common hyperglycemia. Studies with animals have shown that the accumulation of macrophages in diabetic kidneys causes the decline in renal function and contribute to the development of renal injury and sclerosis. The Kefir (K), fermented milk classified as probiotic, composed of a symbiotic mixture of lactic acid bacteria and yeast, showed beneficial health effects such as anti-inflammatory, antioxidant, antiviral, anticancer and antifungal. Furthermore, probiotics have demonstrated the capacity to adhere to mucosal surfaces and inhibit the attachment of other pathogenic bacteria, contributing to the integrity of the barrier, modulating cells of the immune system. The aim is to analyze the effect of Kefir ingestion on the function of peritoneal macrophages and intestinal mucosal from diabetic rats. The DM will be induced in Wistar male adult, by injection of streptozotocin. Upon confirmation of DM animals will be allocated into 4 groups: control (CTL), diabetic (DM), CTLK and DMK treated with Kefir 1.8 mL/day by gavage for eight weeks. After treatment, the animals will be sacrificed in a CO2 chamber and exsanguinated. In resident macrophages, obtained from lavage of the peritoneal cavity will be analyzed parameters of functionality as the phagocytic activity, the ability of adhesion and spreading, in vitro synthesis of cytokines (IL-1 beta, IL-6, IL-10, TNF-alfa, IFN-gama, IL-17 and IL-4), the production of nitric oxide, hydrogen peroxide and superoxide anion. In the intestinal mucosal will be performed histology, enumeration of Peyer patches and flow cytometry of dendritic cells and macrophages. Values will be described as mean ± SEM, and analyzed by one way ANOVA with post-test Newman-Keuls. (AU)
