Abstract

Dental enamel specimens used for in vitro and in situ studies needs to be completely disinfected before being used in research's, however, the enamel properties must not be affected by the sterilization method. The microwave irradiation for 3 min at 650 W result in complete disinfection of enamel and cause no significant changes in the superficial microhardness and on the demineralization and remineralization processes, then, could be considered as a simple and fast alternative to the usual sterilization methods. The aim of this study is to investigate the influence of the microwave irradiation and the other enamel sterilization methods on the enamel properties, in order to find the method that cause less damage on the dental enamel. The evaluated sterilization methods are: I - microwave irradiation, II - Gamma irradiation, III - Ethylene oxide and IV Steam autoclave. This study will be divided into two phases, were the phase 1 will evaluate the effect of the different sterilization methods on the enamel chemical composition (Raman and XPS) and on the enamel morphology (SEM). The phase 2 will evaluate the effect of the different sterilization methods on the early biofilm formation, with adhesion of the biofilm early colonizers (A. naeslundii and S. Sanguis). The evaluated parameters will be: microorganism's adhesion (XTT), total biomass (crystal violet), number of microorganisms (CFU/ ml) and biofilm morphology (confocal microscopy). (AU)
