Amoxicillin efect on the rats enamel development

Abstract

Given the absence of conclusive data about the etiological factors of Molar-Incisor hypomineralization (MIH), the purpose of this study is to evaluate the effect of amoxicillin in the development of dental enamel of rats. The experiment is divided into two phases which will be used in 45 pregnant rats. In the first phase will be a dose-response curve for both, will be used four groups (Ga, Gb, Gc and Gd), with five pregnant rats in each, which will receive a daily dose, intragastrically, amoxicillin from 140 days until 200 days of pregnancy. In Ga: amoxicillin 25mg/kg in Gb: amoxicillin 50mg/kg in Gc: 100mg/kg amoxicillin and Gd: saline solution. This phase will analyze the changes in the enamel of the puppies by histological analysis (H & E), the fluorescence induction analysis using QLF (Quantitative Light-induced fluorescence), and a stereomicroscope (20X magnification). In the second phase, the minimum dose capable of causing enamel found in Phase I will be administered to 25 pregnant rats, which will be divided randomly into 5 groups. GI, 5 rats receive a daily dose, intragastrically, 14 days of amoxicillin for 20 days of pregnancy, the offspring will receive GIA daily dose from 1 to 11 days of life, and Gib receive saline. GII, 5 rats receive daily doses of amoxicillin pregnancy of 14 days to 11 days of life of the offspring, the offspring GIIA receive daily dose of amoxicillin 1 to 11 days of life, and GIIB receive saline. GIII (sham group), 5 rats receive saline from 14 to 20 day of pregnancy, no intervention in the offspring will be held. GIV (control group) in 5 rats no intervention is performed, the offspring Giva receive daily dose of amoxicillin 1 to 11 days of life, and GIVb receive saline. In GV, 5 rats receive daily dose of amoxicillin for 14 days to 20 days of pregnancy, the offspring will be sacrificed on GVA 10 days of life, and fragments of mandible and maxilla, containing the germs of incisors and molars removed and analyzed by histology (H & E) and amelogenin markers. The offspring of the groups I to IV will be sacrificed at 12 days, and part of the 22 days of life, fragments of mandible and maxilla containing the incisors and the molars are removed and analyzed by histologic analysis (H & E) and markers of amelogenin by immunohistochemical reaction. The enamel surface of molars and incisors are analyzed by scanning electron microscopy (SEM) and mineral content will be analyzed by atomic force spectroscopy. The data will be tabulated and analyzed statistically. (AU)
