Abstract

Melanomas are malignant neoplasms that arise from skin melanocytes, the clinical and histopathological features of acral lentiginous melanoma are different from other melanoma types. It is less prevalent in caucasian and more frequent in asian, spanish and african descents. It occurs in the extremities, areas not exposed to ultraviolet radiation, such as palms, soles and subungual region, and is reported to have aggressive behaviour. The authors aim to evaluate the expression of proteins and genes related to the pathogenesis of acral-lentiginous melanoma. Specific objectives: 1) To evaluate the immunohistochemical expression of PTEN protein, c-Kit, cyclin D1, BRAF, Myc, 2) Evaluate the genes CCND1, PTEN, MYC by FISH technique, 3) Validate the FISH diagnostic kit for cutaneous melanoma which includes genes CCND1, RREB1, MYB and centromere 6 in acral melanoma, 4) correlate the immunohistochemical expression of the proteins with gene mutations, 5) correlate the immunohistochemical expression of proteins and gene mutations with pathological data, clinical, and demographic follow up. Patients and Methods: Retrospective data from patients with cutaneous melanoma of the acral lentiginous type, greater than or equal to 1.0 mm, held at the Federal University of São Paulo School of Medicine. The specimens will be obtained from the files of the Department of Pathology and demographic data, clinical and follow-up will be obtained from medical records of these patients. (AU)
