Production of pluripotent stem cells as studied models in dogs

Abstract

Recent trials with animal models have shown the great importance of embryonic pluripotent stem cells for the treatment of various diseases, many of them extremely complex. Regenerative medicine supported by the development of biotechnology emerges with a promising future, one of the most desired therapeutic options. In this context, obtaining stem cell lineages from dog embryos is still a challenge as a model for cell therapy, however, some researchers have shown be possible to isolate these cells from inseminated and castrated females, even though a laborious process. In the other hand, the production of induced pluripotent stem cells (iPS) from dog fibroblasts reached encouraging results for obtaining pluripotent cells and the study of its application in regenerative medicine without severe ethical implications. This proposal is aimed to isolate and cultivate the dog embryo stem cells derived from pre implanted blastocysts, to establish a genetically induced pluripotent stem cells lineage from dogs (iPS - induced pluripotent stem cell), and subsequent characterization of them. It is expected, based on comparative analysis, the iPS produced (in vitro model) require the same conditions of maintenance, and exhibit the same growing behavior as common embryonic stem cells (in vivo model), confirming its applicability from a comparative model of development.
