Evaluation of optical, ultrastructural and expression of cell proliferation of mucosa enteric mucosal broiler supplemented with glutamine and glutamic acid

Abstract

Two experiments will be conducted in poultry sector, Universidade Estadual Paulista "Júlio de Mesquita Filho", in FMVZ / Botucatu, in order to assess the integrity of the intestinal mucosa by means of optical microscopy, scanning electron and transmission electron as well as evaluating the expression of cell proliferation of enterocytes via the technique of mitotic markers PCNA (proliferating cell nuclear antigen), and the performance of broilers supplemented with glutamine, glutamic acid and Aminogut. In the first experiment will be housed 720 chickens a day old, male, distributed in a completely randomized design with four treatments and six replicates of 30 birds each. The treatments are: 1 - Basal Ration (without adding any additives), 2 - basal ration with 1% glutamine, 3 - basal ration with 1% glutamic acid, 4 - basal ration with 2% Aminogut (product association glutamine and glutamic acid), in which the data will be evaluated performance (weight gain, feed intake, feed conversion and viability). In experiment II are housed 340 birds for analysis of intestinal integrity, whose design is entirely randomized with four treatments and five replicates of 17 birds each. The treatment will be the same for both experiments. During the second trial will occur three withdrawals at 7, 14 and 42 days of age, in which the gut and segments of 15 birds per treatment will be weighed and measured with a tape measure. For morphometric analysis of the epithelium will be collected segments of the duodenum, jejunum and ileum, five birds per treatment, which will be opened and washed with distilled water and fixed in formaldehyde solution 10%. Then, the segments will be subjected to routine histological procedures. For the analysis of scanning electron microscopy will be collected segments of the small intestine (duodenum, jejunum and ileum) of five birds per treatment, which will be submitted for routine scanning electron microscopy. For transmission electron microscopy, segments of the duodenum will be collected from eight birds per treatment, which will be submitted for routine transmission electron microscopy. Although these shares will be collected the same segments (duodenum, jejunum and ileum) of five birds per treatment for evaluation of PCNA through mitotic markers, which are subject to routine laboratory analysis of immunohistochemical Veterinary Pathology. The present study aims to evaluate the performance, cell multiplication, as well as the integrity of the intestinal mucosa of broilers supplemented with glutamine, glutamic acid and Aminogut (product association of glutamine and glutamic acid) through Optical Microscopy, Electron Microscopy Scanning and Transmission Electron and Technology of PCNA.
