Abstract

Aneuploidy plays an important role in the pathogenesis of malignant tumors. The oncogene Pituitary tumor Transforming (PTTG) (Pds1/Securin) was described in pituitary tumors. This gene encodes the human protein hSecurin responsible for sequestrating and inactivating the activator of protein separation of sister chromatids called separase. The PTTG is expressed in pituitary tumors,but also described in Adult T-Cell Leukemia/Lymphoma (ATL) associated with virus HTLV-I. As the events that predispose and direct the development of aneuploidy in ATL are still unclear, we set out to study the participation gene hSecurin of the pathogenis ATL. The gene expression hSecurin will be quantified in lymphocytes TCD4+ and TCD8+ of asymptomatic carries of HTLV-I and patients with ATL and a healthy group using the technique polymerase chain reaction (PCR) quantitative Real Time. The gene expression hSecurin will be correlated with index DNA ploidy in lymphocytes TCD4+ and TCD8+ groups of the study, which will be evaluated by flow cytometry. The cases of ATL will be confirmed and differentiated from asymptomatic group of carries of HTLV-I by the detection of monoclonal integration of HTLV-I in malignant cells by Inverse Long PCR technique and a research of rearrangement of T-Cell Receptor (TCR).
