Abstract

Neural differentiation is a multi-stepped process that involves the switching on and off multiple gene expression profiles for the modulation of cell cycle exit and acquisition of early differentiation steps.Recently, our laboratory cloned and characterized the expression pattern of the transcription factor cScratch2 (cScrt2) during chick early posterior neural tube development. This transcription factor belongs to the Snail superfamily of Zinc-finger transcription factors. cScrt2 is specifically expressed neural tube at its early stages of development. In the truncal neural tube, this expression seems to be restricted to recently post-mitotic cells, suggesting that it might have a role in triggering the neural differentiation program.This projects aims to determine the target gents of cScrt2 through deep sequencing of neural tubes that have been electroporated with a dominant negative form of cScrt2 (amino-terminal deleted). After alignment and annotation of the RNASeq transcriptome results, we will prioritize validation of the transcription factors whose expression levels were altered by dominant-negative cScrt2. (AU)
