C-kit mutations analysis in acral lentiginous melanoma

Abstract

Melanoma has its initiation in malignance transformation of melanocytes. Acral lentiginous melanoma (ALM) subtype was once thought to occur more often in more darkly pigmented races and it was also thought to have a more aggressive biologic nature than other subtypes. As the name implies, ALM occurs on acral surfaces, specifically the palms, soles, and nails. Genetics characteristics of this tumors seems to determinate therapeutic efficacy. The c-kit gene codes a tyrosine-kinase receptor whose ligand is the stem cell factor, a growth factor that plays a key role in formation of melanocytes. Mutations in c-kit produce permanent activation without any binding to the ligand and it have been identified in gastrointestinal stromal tumors, in certain leukemias, and in mastocytoses and seminomas. The role of this study is evaluate c-kit mutations in ALM by analysis of tumors different areas. 50 formalin-fixed paraffin-embedded samples from acral lentiginous melanoma will be obtained from the archives of the Department of Pathology, Universidade Federal de São Paulo. Four different areas of each tumor will be selected for DNA extraction. We will evaluate mutation by gene sequencing of exons 11, 13, and 17. Correlation with histopathology and clinical information will occurs as well as possible. (AU)
