Relationship of genotypic characteristics of coagulase-negative Staphylococcus with cure rates after antimicrobial treatment of ovine mastitis

Abstract

Mastitis in sheep producing milk and meat is recognized to affect the milk quality of sheep, which causes economic losses due to less weight gain in the lambs and risks related to the loss of glands or death of the animal. Coagulase-negative Staphylococcus (CNS) are among the main responsible for the disease in sheep, and among the measures to control the disease caused by these pathogen, the treatment at the end of lactation may contribute to the prevention and cure of subclinical cases. However, cure rates can be reduced by various causes, such as those related to virulence factors presented by microorganisms. The objectives of this project are identified in the milk of sheep that will be treated by dry therapy: CNS species before and after treatment; to establish in these micro-organisms the presence of genes mecA, icaA, icaC, icaD, Bap, Bhp, sea, seb, sec, sed and tst-1, related to antimicrobial resistance ²-lactam antibiotics, biofilm formation and toxin production, respectively. Cure rates for spontaneous subclinical mastitis in sheep and cure rates after treatment will be measured and related to the presence/absence of virulence factors in CNS. The sheep will be randomly assigned to three experimental groups: G1 (control), G2 (infusion of 100 mg of cloxacillin), G3 (infusion of 100 mg of cloxacillin nanoparticulate) after prior screening by the California Mastitis Test (CMT) and somatic cell counts (SCC). The results will be confirmed using the microbiological culture. The informations acquired with the project execution will collaborate with farmers and technicians to the best knowledge of the epidemiology of disease caused by CNS and subsequent prevention and control.
