Evaluation of bone repair and mineralization in animals submitted to alendronate treatment

Abstract

The aim of this study is to evaluate, in rats, the effect of bisphosphonate therapy on bone repair process and mineralization around osseointegrated implants. For this, 180 rats will be randomly allocated into two groups: the test group, where the animals will receive subcutaneous administration of alendronate once a week, and the control group, where the animals will receive weekly administration of placebo, during the entire experiment. After 120 days from the beginning of the treatment, a titanium implant will be installed at the tibial metaphysis, of both legs. Ten animals of each group will be sacrificed on days 5, 10, 15, 20, 25, 30, 45, 60 post-surgery. It will be removed the left femur, both tibia and the lumbar vertebrae. Five animals from each group will receive subcutaneous injection of fluorocrômicos markers in order to check the formation of new bone at intervals of 7 days, and will be sacrificed at 30 and 60 days. Blood and urine samples will be collected on the day of sacrifice for evaluation of bone markers. The femur and lumbar vertebrae will be analyzed in order to determine bone mineral density. The tibiae specimens will be submitted to histological, histometric and immunohistochemistry analysis in order to assess the implications of bisphosphonate treatment on bone tissue repair. (AU)
