Abstract

Endodontics in primary teeth is an important procedure to maintain the integrity and health of the teeth and supporting tissues. The use of a root canal filling material that shows desirable properties is indispensable for primary teeth. The aim of the present study is to evaluate the effects of different root filling materials on cytotoxicity from fibroblasts (L929) and osteoblasts (MC3T3.E1) cells lines. The materials will be distributed in five groups: G1 - culture medium without filling material (negative control); G2- zinc oxide/eugenol paste (positive control), G3- Calen® paste, G4- Calen® associated with zinc oxide paste, G5- Calcipex II® paste. The materials will be prepared according to the manufacturer's instructions under aseptic conditions using a vertical laminar air flow system. The cytotoxicity analysis will be evaluated by cellular viability (24, 48, 72 hours) based on MTT reduction test. The negative control group will be considered to have 100% of viable cells. Morphological analysis of the cells will be observed by confocal laser scanning microscopy. Data will be submitted to Kolmogorov-Smirnov, Shapiro-Wilk, ANOVA and Tukey´s tests, at 5% of significance.
