Transcriptomic and proteomic approaches in the study of cardiovascular biomarkers: focus on acute myocardial infarction

Abstract

The main clinical manifestation of atherosclerosis is the acute myocardial infarction (AMI), which is a medical emergency that requires a prompt diagnosis and efficient therapy. The transcriptomic and proteomic approaches are both powerful tools for the study of AMI and may be instrumental to identify news biomarkers involved in the inflammatory and apoptotic process of cardiovascular diseases. The aim of this study is to determine the gene expression (mRNA and protein) pattern in blood cells following an AMI to identify new biomarkers. For our present studies we will select 10 patients with AMI recently submitted to angioplasty and 6 control individuals, all males, age 20-50 years, from the Instituto Dante Pazzanese do Estado de São Paulo, Brazil. Blood samples will be obtained during the first 48h after AMI for biochemical analysis. A global gene expression pattern of circulating mononuclear cells will be determined using DNA microarray chips (CodelinkTM system). The proteomic analysis of plasma will be carried out with using a ProteinChip® Biomarker System, followed by SELDI (Surface Enhanced Laser Desorption/Ionization) mass spectrometry technology for identification and quantification of proteins. Results from this study will enable us to identify changes in gene and protein profile in the plasma, and may be used as a new diagnostic tool for cardiovascular diseases, as well as biomarkers for prevention, therapy control and prognosis of IAM.
