Evaluation of different concentrations of selenium in sheep reproduction

Abstract

This research project aims to investigate the effects of supplementation with different concentrations of Se and interaction between this with Zn, Pb and Mn on reproduction and production of Santa Inês sheep and Dorper. Will be used 30 sheep in pens managed in intensive system which will be divided into five experimental groups (06 animals / group and three animals per breed): control group (GC1) with supplementation of mineral mixture without addition of selenium and supplemented with other same mineral mixture containing 0.2 (G2), 4 (G3), 12 (G4) and 20 (G5) mg Se / kg of mixture. The trial period will be one year and every 49 days will be collected blood samples by puncturing the jugular vein, semen samples by electroejaculation and feces from the rectum, for each experiment will be an adjustment period of 14 days. After harvesting the semen will be analyzed: turbulence, motility, vigor, concentration and morphology, to assess plasma membrane integrity, acrosomal and mitochondrial association is used fluorescent probes and sperm DNA integrity test will be analyzed by the comet, the fructose and citric acid by the technique of Pereira & Janine; Saffran & Densted respectively. Sodium, potassium and calcium will be analyzed by using ion selective. The concentrations of triglycerides, total cholesterol, LDL and HDL in serum will be determined by enzymatic colorimetric method, levels of zinc and manganese in serum, semen, feces and food will be determined by the technique of Atomic Absorption and Flame of selenium and lead by Atomic Absorption Graphite Furnace, moreover, analyze the concentrations of testosterone, progesterone and insulin in serum by commercial radioimmunoassay kits in the solid phase.
