Use of MGIT 960 TB eXiST system as susceptibility test for Mycobacterium abscessus: standardization and evaluation

Abstract

Mycobacterium abscessus is nontuberculous mycobacteria that can cause lung disease similar to tuberculosis, but is also responsible for various types of pulmonary infections and health care associated diseases. This species can live in the environment and the biofilms formation is a strategy for dissemination and resistance to various compounds used for the disinfection of medical and hospital supplies. There is great variability in resistance profile to drugs used to treat this mycobacteriosis and is recommended to perform susceptibility testing (ST) to choose the best therapeutic strategy. So far, there is no automated test approved for M. abscessus ST as no resistance mechanisms and its intervening critical concentrations of drugs to be used in the treatment of mycobacteria were evaluated. This project aims to develop a basic protocol and establish the critical concentrations for the four main drugs in the treatment of disease caused by M. abscessus using MGIT 960 TB eXisT system and evaluate possible phenotypic and molecular interferences.
