Abstract

The genus Leptospira brings pathogenic species, L. interrogans and saprophytic, L. biflexa and intermediates with varying degrees of pathogenicity. It is known that there is a mobile cellular immune response with activation of macrophages expressing genes responsible for the production of cytokines involved in the pathogenesis of leptospirosis during the infectious process. The cytotoxicity has been reported in previous studies that consider the L. interrogans as an inducing agent of apoptosis of macrophages and hepatocytes. However, the mechanism responsible for cell death remains unclear. This study aims to determine the dynamics of the cellular immune response in vitro using cultures of macrophages (J774) when exposed to virulent strains, attenuated saprophytic Leptospira, observed at various times after exposure. To analyze the activation of macrophage cultures in vitro assays to determine the production of reactive nitrogen intermediates (NO), expression of genes for cytokines and interleukins by PCR and the presence and quantify them in the supernatant of cell culture will be performed by ELISA. For these reasons, we seek to verify and compare the kinetics of response of macrophage activity when exposed to Leptospira spp to the pathogenic, attenuated and saprófito inoculum in order to understand the dynamics of immune cell motility.
