Abstract

The genus Leptospira brings pathogenic species, L. interrogans and saprophytic, L. biflexa with varying degrees and intermediate pathogenicity. It is known that during leptospirosis there is a mobile cellular immune response with activation of macrophages expressing genes responsible for the production of cytokines that participate in the pathogenesis of the infection. The cytotoxicity has been reported in previous studies that consider the L. interrogans as an inducer of apoptosis in macrophages and hepatocytes. However, the mechanism responsible for cell death remains unclear. This study aims to determine the dynamics of the cellular immune response in vitro using macrophage cultures (J774) when exposed to virulent strains, attenuated and saprophytic Leptospira, observed at various times after exposure. To analyze the activation of macrophages in vitro cultivation will be carried out tests which determine the production of reactive nitrogen intermediates (NO) and quantification of cytokines in the cell culture supernatant using the ELISA test. For these reasons, it seeks to check and compare the response kinetics of macrophage activity when exposed to Leptospira spp, against pathogenic inoculum, attenuated and saprófito in order to understand the dynamics of cellular immune mobility.
