Tumor microenvironment in indolent non-Hodgkin lymphomas: role of tumoral microRNAs and inflammatory polymorphisms in clinical and pathological aspects

Abstract

Indolent lymphomas (IL) represent a particular group among non-Hodgkin lymphomas (NHLs) that do not achieve complete response to therapy, which results in relapses. The inflammatory tumor microenvironment and tumoral microRNAs have been recently considered in progression of these tumors. This study aims to describe a detailed composition of the tumor microenvironment of mantle cell lymphoma and follicular lymphoma (MCL and FL, respectively) obtained from patients treated at the Blood Center at UNICAMP. In parallel, genetic polymorphisms in the immunity genes IL2, IL12A, IL10, TGFB1, TGFBR1, TGFBR2, IL17A and IL17F will be studied in each patient, which may interfere with the composition of the microenvironment cells and individual clinical course. MicroRNAs from tumor tissues will also be studied, in order to assess influence in the clinical outcome and in the composition of tumoral microenvironment. The inflammatory cells and the cytokines encoded by each gene here considered will be assessed by immunohistochemistry, and genotyping will be performed by the Openarray" system. The microRNAs will be studied by means of next-generation sequencing, performed by the Illumina® platform. Disease progression will be achieved by review of medical records. We hope that this study will contribute to characterize distinct groups of patients with MCL and FL, expanding the knowledge about their pathophysiology and enabling the development of more reliable prognostic markers for these diseases, in the context of personalized medicine.
