Abstract

The objective of this project is related to the mechanism of drug action from the pharmacological-chemical, molecular, and therapeutic points of view. The chosen drugs and methods might change the drug-receptor system on hypertension, hyperglycemia, and modulation by histamine. Subproject I: Study of cellular homeostasis of calcium in peripheral neurotransmission in cardiovascular and metabolic diseases. The objective is to: 1- To study the mechanisms the can lead to a sympathetic disfunction, analyzing mainly the release of catecholamines stimulated with angiotensin II. 2- To quantify the concentration of ATP in chromafin cells. 3- To study functional changes in isolated atrium, considering chronotropism and inotropism. 4- To study functional sympathetic changes, considering purines in isolate atrium considering chronotropism and inotropism. 5- Release of catecholamines stimulated by purinegic agonists. 6- To study cellular homeostasis of calcium. Subproject II: Modulatory effect of histamin in isolated rat testis capsule. Specific objectives: 1- Evaluate pharmacologically which are the histamine receptors involved in the modulation of sympathetic neurotransmission. 2- Analyze, through molecular biology, the expression of the level of transcripts for histamine receptors. 3- Verify if stored histamine is released from sympathetic peripheral terminals. 4- To study the interaction of postsynaptic responses induced by histamine. 5- To study the interaction of adrenergic and histaminergic auto receptors. 6- Evaluate the effect of histamine on maturation and of transit time of spermatozoids.
