Abstract

Although there are already clinical studies in humans to evaluate the potential of the use of mesenchymal stem cells (MSC) for cell therapy, it is still necessary to define the best protocols to obtain cells in quantities and qualities satisfactory for use in cell therapies. DNA methylation is one of the most studied epigenetic events, and it represents an important pathway for gene expression regulation in eukaryotes during the development process, in the cellular phenotypes definition, as well as participates in the initiation and progression of several diseases. Recently, the use of tools such as next-generation sequencing (NGS) has allowed investigating, global and punctually, how DNA methylation affects the different biological systems. Thus, the main objective of the this project is to study in large-scale (NGS) the profile of DNA methylation of the MSC derived from human dental pulp (DPSCs). DPSCs will be cultured with platelet lysate, which actually have being suggested, as viable alternatives for the MSCS expansion for cell therapy in humans, but still need further validations. The proposal execution could provide relevant informations DPCs expasion for cell therapy and it can give sequence and consolidate the establishment of tool for large-scale analysis of DNA methylation and mRNA expression in the Department of Morphology from the FOP - UNICAMP, where there is already projects with epigenetic studies related to immuno-inflammatory diseases.
