Insulin and glucagon in the acute inflamation by Aeromonas hydrophila in Nile Tilapia Oreochromis niloticus

Abstract

There is clear evidence demonstrating the involvement of the endocrine system in the inflammatory process. Although its importance, there is lack of literature about the interaction of the inflammatory process with pancreatic hormones in fish. Therefore, the aim of this study is to evaluate the effect of insulin and glucagon in acute inflammation induced by inactivated Aeromonas hydrophila in Oreochromis niloticus (Nile tilapia). For this, 960 Nile tilapia (± 200g) will be divided into 8 groups (3 repetitions): T1: (control group) - treated with sterile saline (0.65%), T2: treated with 30 mg/kg of metyrapone, T3: treated with 30 mg/kg of metyrapone + 0.75 mg/kg of glucagon, T4: treated with 30 mg/kg of metyrapone + 0.8 mg/kg of cortisol, T5: treated with 30 mg/kg of metyrapone + 0.75 mg/kg of glucagon + 0.8 mg/kg of cortisol, T6: treated with 2 IU of insulin, T7: treated with 40 mg/kg of alloxan, and T8: treated with 40 mg/kg of alloxan + 2 IU of insulin. All treatments will be composed of a group challenged with inactivated A. hydrophila and a control group injected with saline. The animals will be euthanized at the times 30 min, 1, 3, 6, and 24 hours after challenge with the bacteria in the swim bladder to collect the samples. The animals will be evaluated for their clinical behavior, inflammatory cell component, hematologic variables, serum biochemistry, cortisolemia, blood glucose, determination and quantification of the cytokines TNF, IL-1 and IL-8 in serum, respiratory activity of leukocytes, concentration of lysozyme, lytic activity of serum, bacterial agglutination, complement system, hemolytic activity, histopathology, and percentage of relative survival.
