Abstract

Osteoarthritis (OA) is a chronic degenerative process characterized by progressive deterioration of the cartilage, subchondral bone remodeling, and loss of joint space, osteófitosis, and loss of function. Current Interventions for OA have as main objectives alleviate symptoms, reduce pain and inflammation control, however these pharmacological interventions and physical therapy has little or no effect on the repair of articular cartilage. Mesenchymal stem cells (MSCs) are being considered for the treatment of a wide range of diseases, and researchers are particularly interested in their potential for the treatment of musculoskeletal disorders, such as osteoarthritis, osteonecrosis, and osteoporosis. However there are still issues related to bioavailability and adhesion of these cells into the joint space. The low-level laser therapy (LLLT) has been shown to promote proliferation of various cell types, including mesenchymal stem cells. However the better understanding of the use of laser therapy in preconditioning and post-implantation of MSCs irradiation shows up required. In this study will be used MSCs extracted from adipose tissue of rats (r MSCs) and pre conditioned with LBI and not conditioned cells and irradiated after intervention by direct injection into the joint space of knee isogenic rats subjected to experimental protocol OA. Given the above this study aims to determine if laser therapy of low intensity can provide increased bioavailability of mesenchymal stem cells injected into knee rats undergoing experimental model of OA, improving chondroprotective benefits, reducing inflammation and the expression of pro-inflammatory cytokines such as IL-1 beta, IL-6 and TNF-alpha, matrix metalloproteinases (MMPs), degradation of proteoglycans in cartilage.
